Differents methods to the determination of nitrate resductase activity in Lotus sp. tissues

Nitrate reductase activity (NRA) from stem and leaves of Lotus corniculatus and Lotus tenuis was assayed in vivo, in situ and in vitro. There was a strong correlation comparing assays in vivo with and without added nitrate and between in situ and in vivo without added nitrate. The highest absolute values of NRA in all tissues assayed where obtained using the in vivo with added nitrate methodology. The advantages and disadvantages of the differents methods in the estimation of NRA for birdsfoot trefoil are discussed.
